Crab meat and roe are highly nutritious delicacies in China. While extensive research has been conducted for allergens derived from crab-meat, data relevant to the allergenic potential of crab roe derived proteins, of which hemocyanin is a principal contender, are almost entirely absent. Using bioinformatics prediction and IgE-binding assays, the three principal immunodominant epitopes of hemocyanin were identified and then combined as a single recombinant fusion protein (rHc). This together with the full-length recombinant protein (Hc) were expressed in Escherichia coli and subsequently identified by SDS-PAGE and immunoblotting. Ninety-five percent of our patients were found to carry rHc-specific IgE antibodies by ELISA. Dot-blot inhibition, together with ELISA inhibition studies, showed that pre-incubation of patient sera with the recombinant epitope protein could inhibit26% to 63% (mean: 50%) of IgE binding to immobilized, full-length Hc and the dose-response curve represents as a sigmoid shape. The recombinant protein (rHc) represents a versatile biologic tool with which to diagnose and investigate therapies for E. sinensis allergy.  相似文献

The misfit dislocations and strain fields at a Ge/Si heterostructure interface were investigated experimentally using a combination of high-resolution transmission electron microscopy and quantitative electron micrograph analysis methods. The type of misfit dislocation at the interface was determined to be 60° dislocation and 90° full-edge dislocation. The full-field strains at the Ge/Si heterostructure interface were mapped by using the geometric phase analysis (GPA) and peak pairs analysis (PPA), respectively. The effect of the mask size on the GPA and PPA results was analyzed in detail. For comparison, the theoretical strain fields of the misfit dislocations were also calculated by the Peierls-Nabarro and Foreman dislocation models. The results showed that the optimal mask sizes in GPA and PPA were approximately three tenths and one-tenth of the reciprocal lattice vector, respectively. The Foreman dislocation model with an alterable factor a = 4 can best describe the strain field of the misfit dislocation at the Ge/Si heterostructure interface.  相似文献

Three kinds of fish from a natural fishpond in the north of Taiwan were collected and analyzed in this study. The three species were non-predatory Ctenopharyn odon idellus, predatory Aristichthys nobilis and predatory Mylopharyngodon piceus. The activity concentrations of 90Sr in bone and edible flesh of fish, as well as in water and soil of fishpond were measured by using the radiochemical analysis. Additionally, the concentration of stable calcium in fish bone, [Ca]bone, and that of stable strontium in fish bone, [Sr]bone, were measured by ICP-AES. From the results, the concentration factors of 90Sr, CF(90Sr), in fish bone was no statistic difference between non-predatory and predatory fish. Besides, the accumulation of 90Sr in the non-predatory fish remarkably decreased with increasing the fish weight. As for the predatory fish, they both showed no statistically significant correlations between the 90Sr activity concentration and the fish weight. Regarding the activity concentrations of 90Sr in fish bone, all the fish were observed positively correlated with the measured [Sr]bone.  相似文献

Oxalic acid, which is one of the most common dicarboxylic acids, is expected to be an important component of atmospheric aerosols. However, the contribution of oxalic acid to the generation of new particles is still poorly understood. In this study, the structural characteristics and thermodynamics of (C2H2O4)(CH3NH2)n (n = 1–4) were investigated at the PW91PW91/6-311++G(3df,3pd) level of theory. We found that clusters formed by oxalic acid and methylamine are relatively stable, and the more the atoms participating in the formation of a ring-like structure, the more stable is the cluster. In addition, via the analysis of atmospheric relevance, it can be revealed that clusters of (C2H2O4)(CH3NH2)n (n = 1–4) have a noteworthy concentration in the atmosphere, which indicates that these clusters could be participating in new particle formation. Moreover, by comparison with (H2C2O4)(NH3)n (n = 1–6) species, it can be seen that oxalic acid is more readily bound to methylamine than to ammonia, which promotes nucleation or new particle formation. Finally, the Rayleigh scattering properties of clusters of (C2H2O4)(CH3NH2)n (n = 1–4) were investigated for the first time to determine their atmospheric implications.

Treatment of N,N-dimethylacetamide (DMAC) wastewater is an important step in achieving the sustainable industrial application of DMAC as an organic solvent. This is the first time that treatment of a high concentration of DMAC in real wastewater has been assessed using membrane bioreactor technology. In this study, an anoxic–oxic membrane bioreactor (MBR) was operated over a month to mineralize concentrated DMAC wastewater. Severe membrane fouling occurred during the short-term operation of the MBR as the membrane flux decreased from 11.52 to 5.28 L (m2 h)−1. The membrane fouling was aggravated by the increased amount of protein fractions present in the MBR mixed liquor. Moreover, results from the excitation–emission matrix analysis identified tryptophan and other protein-like related substances as the major membrane-fouling components. Furthermore, analysis of the DMAC degradation mechanism via high performance liquid chromatography (HPLC) and ion chromatography (IC) revealed that the major degradation products were ammonium and dimethylamine (DMA). Although the MBR system achieved the steady removal of DMAC and chemical oxygen demand (COD) by up to 98% and 80%, respectively at DMAC0 ≤ 7548 mg L−1, DMA was found to have accumulated in the treated effluent. Our investigation provides insight into the prospect and challenges of using MBR systems for DMAC wastewater degradation.
